Situated in the historic town of Osuna, in the province of Seville, the college was founded in 1548 by the Order of the Purísima Concepción, with the aim of offering both religious and academic education. The origins of the Colegio-Universidad de la Purísima Concepción date back to the mid-16th century, when, under the patronage of the local nobility and the initiative of the Order of the Purísima Concepción, it was established to provide religious education to young people in Osuna and the surrounding areas. The foundation was led by Francisco Téllez-Girón, the Cid of Osuna, with the goal of creating an educational institution to rival the great schools of the time.

Over the years, it has educated several important figures in politics, culture, and science, many of whom played crucial roles in the history of the town and Andalusia.

The influence of the university was significant, not only in the formation of intellectual elites but also in the development of educational infrastructure, which contributed to the cultural growth of Osuna and its surroundings. The University of the Purísima Concepción remained active until the early 19th century, offering quality academic programs in various fields and adapting to the changing social, economic, and cultural dynamics of the time.
